Ms. Chalice comes with her own set of special moves compared to the powers of Mugman and Cuphead. One of the best parts of her addition is that she is not just a side character. You can play with her from the start until the end. How To Unlock Ms. Chalice In Cuphead: The Delicious Last Course
You will need to start by beginning the DLC quest for Cuphead. This will require you to head to a new Island. It will be a simple journey to the first couple of levels during which you will also see some cutscenes. Once you are done with the introduction, you will gain access to an item called the Astral Cookie.
you will need to equip this Astral Cookie in order to unlock Ms. Chalice as a playable character in the game. This will take up a slot for the Charm on either Mugman or Cuphead so decide for yourself how you will tackle it.
